The Access Board sets Rehab Act § 508 & ADA standards — i.e. regulations
and guidelines.

They're a tiny specialist agency, sorta like the OGIS is for FOIA. Setting
accessibility standards & guidelines (and consulting on how to implement
them) is more or less literally all they do.

Their standards are mandatory for all Federal agencies, but mainly apply at
the time of acquisition or building new stuff — there's usually some sort
of grandfather clause to allow continued use of current equipment. I don't
recall offhand if the standards are also mandatory for private
organizations taking Federal money (i.e. Rehab Act) or for ADA in general.
