You can do either.  I find it stays on better using peg 1.  When you use peg
1, it counts as your first stitch and you can knit it off just like an
e-wrap stitch.

Good morning everyone!

For those of you that use knitting looms, I have a question! When you cast
on using the e-wrap method, do you put your slip knot on the anchor peg or
on one of the pegs of the loom? The Loom Knitting Primer book says you can
do either, but if you put it on one of the pegs on the loom, does this count
as the first loop on that peg? 

Also, do you all have a preference between going around the loom clockwise
or counterclockwise?
