Well, first of all, I would see the dog has been relieved right before you fly. If you think he will need to go out during the lay over, then ask for assistance when you are exiting the plane. You can probably ask for it in advance. You will probably have to leave the secure area and will have to go through security again, but that may depend on the airport. Experiences in hotels vary widely. Some have areas to relieve the dog, meaning 
someplace where the dog will go. I don't know what your dog's preference is. Mine will use grass or concrete, or areas around trees. Often if you are seeking a place, though, the bell staff is willing to be of help, so just make your needs known. I'd try to keep the dog on as much of a routine as you can, but it is hard to give you much help not knowing the situation. I travel all the time, so I guess I just kind of do it. It isn't really too difficult though, and I think you will enjoy it, specially if the dog loves to fly like mine does.
